Cell dose is an important parameter in predicting a good outcome in cord blood transplantation; particularly, the extent of functional progenitor cells infused. Classical tests use the approach of testing viability by nuclear dye exclusion; however, these only detect latest stages of cellular death. Colony Forming Units (CFU) is the most commonly used functional assay but is difficult to standardize and requires 7-14 days for accurate results, and, therefore, is not practical as a releasing test. We proposed the use of flow cytometry to evaluate the functional dose by using early apoptosis to predict CFU and thereafter be used as a fast releasing test in cord blood banking. This methodology was selected as it represented one of the simplest and robust methods for assessing the cell dose, but it is not the only method. This webinar will address the various methods for progenitor cell assessment that have been used and their applicability in the CB field.
